College of Marine Life Sciences,Ocean University of Qingdao,. 青岛海洋大学海洋生命学院,山东,青岛,266003 青岛海洋大学海洋生命学院,山东,青岛,266003 报道了从食草性海螺粒蝾螺和锈凹螺中制备海螺酶的方法,并且初步研究了酶的组分、性质及其在海藻生产上的应用.酶学分析表明:两种海螺酶均为复合酶,含有纤维素酶、木聚糖酶、褐藻酸酶、几丁质酶、α-和β-半乳糖苷酶和β-葡萄糖苷酶;同时研究也表明除β-葡萄糖苷酶、α-和β-半乳糖苷酶外,大部分酶的最适pH在6-7之间.用复合酶Ⅰ(粒蝾螺酶)和复合酶Ⅱ(锈凹螺酶)溶液分别水解条斑紫菜、海带组织,均获得大量单细胞和原生质体.将获得的单细胞在改进的PES培养基中培养,细胞能正常分裂、长成正常的植株.在实际生产上,将获得的大量紫菜单细胞附着于尼龙网上,经过实验室短期培养再移于海中可长成正常叶状体,初步表明该方法在实际生产上具有广阔的应用前景. Two complex enzymes were extracted from, respectively, Turbocoronatus granulatus(gme-lin and Chorostoma rastia. The activity and properties these enzymes were determined. The results showed that these complex enzymes were consisted of cellulose, xylanase, alginase, chtinase, α-(β)galactosidase, β-glucosidase and protease. Optimum pH of these enzymes were about 6 to 7, except α-(β)galactosidase and β-glucosidase. The amount of single cells and protoplasts were obtained after laver were treated by 1.0% mixed enzyme Ⅰ at 32℃, pH6. 6 and kelp were treated by 1.0% mixed enzyme Ⅱ at 22℃ pH7. 0 respectively. The single cell cultured in PES medium began to meiosis and became normal thallus. Most of laver single cells can adhere on nylon net and become normal thallus in the sea after temporally culture at lab. The single cell of kelp begins meiosis after 15 days and become filament 20 days later.
